- This differential staining is used for identification of Mycobacteria and Nocardia species.
- These microbes contain tough lipid cell wall, which is difficult to stain with ordinary stain.
- Strong stain is used with application of heat that allows stain to enter in the cell.
- Acid fast bacteria does not decolourized by Acid fast decolourizer and stains red whereas, non acid fast decolorizes and stain with counter stain methylene blue as blue.
